The Large-scale Deletions and Copy Numbers of Mitochondrial DNA in Gout Patients Treated with Allopurinol

چکیده

Allopurinol is now the first-line treatment for managing gout and its increasing usage has brought more patients at risk of developing allopurinol-induced hypersensitive reactions. Up to now, our attempts identify biomarkers hypersensitivity allopurinol are still finite. Besides, association between drug-induced reactions some mitochondrial DNA (mtDNA) alterations been proved but remains unclear in patients. In this study, 41 blood samples prescribed with including 22 29 tolerant ones were used analyze their mtDNA large-scale deletions through PCR determine deletion level copy number by quantitative PCR. The results indicated that group lower than one (p < 0.05). However, no was found appearance addition, study showed negatively correlated numbers (R = -0.3495; p 0.0119) -0.6744; 0.0005). Thus, a decrease might serve as potential biomarker further investigation assess patients.

Resistance training in patients with single, large-scale deletions of mitochondrial DNA.

Dramatic tissue variation in mitochondrial heteroplasmy has been found to exist in patients with sporadic mitochondrial DNA (mtDNA) mutations. Despite high abundance in mature skeletal muscle, levels of the causative mutation are low or undetectable in satellite cells. The activation of these typically quiescent mitotic cells and subsequent shifting of wild-type mtDNA templates to mature muscle...

Disease progression in patients with single, large-scale mitochondrial DNA deletions

Single, large-scale deletions of mitochondrial DNA are a common cause of mitochondrial disease and cause a broad phenotypic spectrum ranging from mild myopathy to devastating multi-system syndromes such as Kearns-Sayre syndrome. Studies to date have been inconsistent on the value of putative predictors of clinical phenotype and disease progression such as mutation load and the size or location ...

Withdrawal of allopurinol in patients with gout.

The xanthine oxidase inhibitor allopurinol effectively lowers the plasma level of uric acid and the drug has become widely used in the treatment of gout, especially in patients with renal failure, uric acid calculi, or intolerance to uricosuric agents (Scott, Hall, and Grahame, 1966). Various theoretical complications of treatment have been considered from time to time, such as possible incorpo...
